It was a nail-biting finish to the race as the GB duo were pushed all the way to the line in a time of 6 minutes 22.47 behind Lithania and Slovenia.
Townsend and Lucas, racing in their first Olympics, took the third qualifying spot and will contest the final on Thursday at 11.50am.
Also into the final in impressive fashion are the lightweight men's four, who won their semi-final in 5:59.68.
Rob Williams, of Maidenhead, who learnt to row on Dorney Lake, will race in Thursday's final with crewmates Peter and Richard Chambers and Chris Bartley.
Switzerland were their main rivals in the race, but a great sprint finish in the last 500m saw GB over the line in first place with Netherlands coming third.
